Studies on the Production of Yeast

Abstract
During the course of investigation on the production of RNA, it has been found that the RNA content was in wide distribution in various kinds of yeasts, and Cryptococcus laurentii had especially high RNA content and high dry cell weight.
Time courses of the RNA synthesis and dry cell weight of Cryptococcus laurentii were examined, and the most desirable results were obtained at 48 hours of cultivation on a rotary shaker at 30¡É under the pH 5.6. Under the above condition, the RNA content and the yield of dry cells were examined changing the medium composition in various ways.
Glucose was observed favorable as a carbon source, and yeast extract and meat extract as a nitrogen source. The optimum C/N ratio of the medium was 8 : 1. The optimum concentrations of KH©üPO©þ, NaC1 and MgSO©þ¡¤ 7H©üO were 0.1 %, 0.4% and 0.1 %, respectively.
Glutamic acid mother liquor, Zn ion or Al ion was added to the medium. Addition of Al ion by 50 ppm showed relatively favorable effects as compared with that of the other materials in increasing the RNA content. Comparing with the basal medium, the RNA content on dry basis was increased from 18% to 23% in the final medium, whereas the yield of dry cells maintained a high amount at the level of 26 to 28 mg per ml of culture broth.
